When setting up your Marketing Cloud Connect integration, completing the Update Page Layouts steps results in a 'Marketing Cloud Unsubscribe' or 'Marketing Cloud Resubscribe' link being added to the page layout of your Contact and Lead records. However, if your Sales and Service Cloud user does not have the required permissions or is not integrated with a user in Marketing Cloud, clicking these links can result in an alert displaying with the error message 'You must be connected with Marketing Cloud to update an Marketing Cloud subscriber.'
Solución
To resolve this error confirm that you have applied all the required permissions to the Sales and Service Cloud user, which are outlined in Marketing Cloud Connect User Types and Permissions.

For example, this specific error has been observed when the Marketing Cloud for Appexchange User permission set is not assigned to the user. In Sales and Service Cloud, navigate to Setup and search 'Users' in the Quick Find. In Lightning Experience, select Users > Users under the Administrator section. In Salesforce Classic, select Manage Users > Users under the Administer section. Find the username of the Sales and Service Cloud user who is encountering the error. Click into the user record and confirm if the Marketing Cloud for Appexchange User checkbox is enabled.

This specific error also occurs if you have not integrated a Marketing Cloud user with the Sales and Service Cloud who is experiencing the issue. This can be achieved by following the steps in Manage User where the Sales and Service Cloud username is inputted when you click Integrate under Salesforce.com Status within the Marketing Cloud user record.
